Abstract:Objective: To investigate the relation between the cytoplasmic region of each subunit of leukemia inhibitory factor(LIF) receptor and cell proliferation related gene, c myc expression level in human leukemia line Meg 01. Methods: We prepared the chimerical receptor exchanged cytoplasmic domain (190/130, 130/190) and separately expressed it on the membrane of Meg 01, then competitively bound LIF with the wild receptor. Therefore, the cytoplasmic region homodimerization (190cyt 190cyt, 130cyt 130cyt) initiated the intracellular signal transduction,the cell proliferation and differentiation. c myc expression level was observed by means of immunoblotting and immunohistochemistry in Meg 01. Results: Higher level of c myc and the faster cell proliferation were determined in group 190/130 compared with the wild type receptor subunit. Conclusion: The cytoplasmic domain of gp130 involves in the induction of c myc expression and the cell proliferation of Meg 01.
