胶原/细菌纤维素复合材料的细胞相容性评价

摘    要:目的评价胶原/细菌纤维素复合材料的细胞相容性。方法将胶原(collagen,Col)和细菌纤维素(bacterial cellulose,BC)通过物理交联法和化学交联法制备成Col/BC物理组和Col/BC化学组,以BC为对照组,采用SEM(扫描电镜)观察材料形貌。采用小鼠胚胎成骨细胞株(MC3T3-E1)与材料复合培养,分为空白对照组(等量的细胞悬液)、材料对照组:BC、实验组1:Col/BC物理组、实验组2:Col/BC化学组观察细胞生长状况,MTT(四氮唑盐)观测细胞在1、2、3、4、5、6 d后的增殖情况,细胞与材料复合第48 h,SEM观察细胞粘附形貌。结果 SEM观察在BC纤维上均匀包覆了一层蛋白质,MTT检测结果表明3 d后Col/BC(物理交联组)组高于其他组,实验组与材料对照组:BC相比,差异有统计学意义(P<0.05)。SEM观察在细胞与材料复合培养48 h,纤维表面均有少量成骨细胞粘附。结论与材料对照组:BC相比,Col/BC物理组和Col/BC化学组均具有良好的细胞相容性,其中,Col/BC物理组表现出更好的细胞相容性。

Abstract:Objective To evaluate the cell compatibility of collagen/bacterial cellulose composites.Methods The Col/BC composites were prepared by physical and chemical cross linking of Col and BC,where BC was assigned as the control group and SEM was used for morphological observation.The embryonic osteoblast cells of mice are co-cultured with the BC,Col/BC physical group,Col/BC chemical group and control group,respectively,and examined by MTT and SEM.Results SEM showed that the bacterial cellilose was uniformly coated with protein.MTT showed that the Col/BC physical group was better than the other groups after 3 days.SEM observed adhesion of a few cells.Conclusions Both Col/BC composites prepared by physical and chemical cross linking show good cytocompatibilty,and the physically prepared is even better.
