硫代硫酸钠减轻豚鼠顺氨氯铂耳毒作用的电生理和组织学研究

摘    要:本实验以内耳生物电及耳蜗浦片为指标,观察硫代硫酸钠对顺氨氯铂(CDDP)耳中毒预防。实验豚鼠分三组,各10只。对照组注射生理盐水,阳性对照注射CDDP2mg/kg;预防组注射硫代硫酸钠800mg/kg,同时给予CDDP2mg/kg;三组均每日一次腹腔给药,共6d。结果:①耳内生物电0~80dB不同程度短声引起的耳蜗微音器电位与听神经动作电位幅值,预防组动物均高于阳性对照组,但两者均低于对照组;②耳蜗铺片,阳性对照组大部分动物耳蜗各回的毛细胞严重性缺损,预防组耳蜗病变仅局限在底回。结果表明硫代硫酸钠能减轻CDDP的耳毒性,为CDDP致聋的防治提供了一条新的研究途径。

Abstract:The bioelectric response of the inner ear and surface preparation of the cochea were chosen as the indicators and the preventive effect of Na2S2O3 on CDDP ototo-xicity were observed in Guinea-pings. Animals were divided three groups per group was ten. Guinea-pigs in contral group normal saline solution wrs injectied. n GDDP group CDDP (2mg/kg) were injected. In Na2S2O3 group Na2S2O3 (800mg/kg) and CDDP (2mg/kg) were injected. All groups were adminitered onoe daily for 6 clays. Results show: (1) The amplitude of CM and AP elicited by the clicks at the intensities from 0 to 80 db in Na2S2O3 group were higher than. that in ODDP group but both of them were lower than that in control group. (2) Damage of the hair cells in CDDP group was sevious while only slight degeneration of the hair cells occurred in the basal coil in Na2S2O3 group. (3) According preceding results the sodium thiosulfate is capable of relieving to a certain extent the ototoxioity of ODDP. It provides new studies way for the preventive effector ODDP ototoxioity.
