| 摘 要： | 利用低渗透析-Sephadex G200层析制备电泳纯、并具免疫活性的IgM型抗Pv红内期McAb(6H_6G_(11))免疫家兔,获得抗-6H_7G_(11)独特型抗体(Anti-6H_7G_(11)Id)。经免疫双扩散、ELISA以及竞争性免疫实验和动物实验表明,该Anti-6H_7G_(11) Id具有针对相应独特型决定簇的特异性,为半抗原抑制性Ab_2,并能模拟Pv抗原的作用,为交叉反应独特型。

| Abstract: | Through hypotonic dialyzingand gel filtration on Sephadex G200, 6H_7G_(11)McAb-IgM (Mouse IgM) was successfullypurified. Anti-6H_7G_(11)Id (Ab_2) was inducedin rabbit. This Ab_2 was identified by agardiffusion, ELISA and competitive assays. Theresults showed that the Ab_2 was able tocombine respective Ab_1 (6H_7G_11 McAb), andthe binding between Ab_2 and Ab_1 could beinhibited by Plasmodium vivax (P.v) anti-gen. Meantime, by using Ab_2 thus preparedto immunize BALB/c mice, the anti-ati-6H_7G_(11)Id (Ab_3 or Ab_1) was obtained withspecificity of Ab_1. It was revealed that theAb_2 thus prepared could mimic the antigenicdeterminant of P.v, and it belongs to thecross-reacted idiotype. |
