红栓菌胞外多糖分离纯化和性质研究

摘    要:目的:分离并研究红栓菌胞外多糖的结构性质。方法:红栓菌发酵液,滤去菌丝等固形物,浓缩发酵液至1/4原体积,经乙醇沉淀、脱色、除蛋白、乙醇沉淀、干燥得多糖粗品PPS1,PPS1经DEAE-纤维素DE-52以及Sephadex G-150纯化得多糖纯品PPS2;采用凝胶过滤、醋酸纤维膜电泳、红外光谱、紫外光谱、核磁共振谱、气相色谱等方法测定多糖的性质。结论:从红栓菌中获得的多糖PPS2为单一组分,相对分子量为29200,由木糖、甘露糖、葡萄糖和半乳糖等单糖组成。

Purification and Characterization of Polysaccharide from Pycnoporus cinnabarirus
Abstract:OBJECTIVE:To isolate and study the polysaccharide from Pycnoporus cinnabarirus METHOD:The fermentative fluid was concentrated to 1/4 volume, precipitated by ethanol, decolorized and deproteinized by Sevag method, then a crude polysaccharide PPS 1, was obtained. After purification by ionexchange chromatography on DEAE cellulose DE 52 and molecular sieve chromatography on Seperdex G 150, the refined polysaccharide from PPS 1, named PPS 2, was obtained. PPS 2 was characterized by means of gel permeation chromatography, cellulose acetate membrane electrophoresis, IR, UV, 1 H NMR and gas chromatography. RESULT: PPS 2 was shown to be homogeneous, the relative molecular weight was estimated to be approximately 29 200,and the component monosaccharides were D xylose, D mannose, D glucose and D galactose.
