针灸对克罗恩病大鼠P物质与神经激肽1受体表达的影响

摘    要:目的观察克罗恩病模型大鼠结肠黏膜中P物质(SP)和神经激肽1受体(NK-1R)表达的变化,以及隔药灸与电针治疗对其的影响。方法除正常组外,其余大鼠采用TNBS灌肠法制备克罗恩病模型,在确定模型成功的基础上,随机分为模型组、隔药灸组与电针组;隔药灸组与电针组分别予以隔药灸、电针“天枢”、“气海”治疗。采用免疫组化方法检测各组大鼠结肠组织SP及其受体NK-1R的表达。结果与正常组大鼠比较,模型大鼠结肠SP、NK—1R表达异常升高.隔药灸与电针对其有显著下调作用,尤以隔药灸作用显著,基本恢复正常水平。结论针灸治疗可能是通过下调SP、NK-1R的表达。消除肠道炎症,改善组织损伤,发挥治疗作用。

Abstract:Objective This paper studied the effects of acupuncture and moxibustion on substance P(SP) and neurokinin-1 receptor(NK-1R) in Crohn's disease(CD) rats. Methods The CD rats model was established by TNBS clusis. When the model was established successfully, SD rats were randomly divided into CD model group, herbs-partition moxibustion(HPM) group and electro-acupuncture(EA) group. The HPM group and EA group rats were treated by herbs-partition moxibustion and electro-acupuncture respectively at Tianshu(ST 25, bilateral) and Qihai(CV6). SP and NK-1R expressions in colonic mucous membrane were detected by immunohistochemistry. Results Compared with the normal rats, SP and NK-1R expressions increased in CD rats' colonic mucous membrane,and were markedly down-regulated by herbs-partition moxibustion and electro-acupuncture treatment. In HPM group, SP and NK-1R expressions recovered to normal. Conclusions Herbs-partition moxibustion and electro-acupuncture treatment may be therapeutically beneficial to CD rats by down-regulating SP and NK-1R expressions.
