光敏剂PSD-007对人骨肉瘤MG-63细胞体外光动力作用的实验观察

摘    要:目的观察PSD-007对人骨肉瘤MG-63细胞的光动力效应。方法以人骨肉瘤MG-63细胞为研究对象,测定以PSD-007作为光敏剂,630 nm波长激光为光源,不同能量密度激光的PDT处理后MG-63细胞的OD492值,并绘制MG-63细胞PDT后的存活率曲线,光镜下观察PDT后MG-63细胞形态的变化。结果 PSD-007浓度为2μg/ml,激光能量密度为6 J/cm2时,PDT处理后MG-63细胞存活率明显降低,仅为48.09%。PDT后MG-63细胞形态发生明显的变化,部分细胞坏死。结论 PSD-007对人骨肉瘤MG-63细胞具有明显的抑制作用。

Abstract:Objective To observe the photochemotherapeutic effect of PSD-007 on MG-63 cell in vitro.Methods Human osteosarcoma cell line MG-63 as the research object,PSD-007 as the photosensitizer,and 630 nm wavelength laser as the light source,The MG-63 cells treated by different concentrations of PSD-007 were treated by different doses of laser irradiation.After the irradiation,MTT assay was used to measure the OD492 values of them,then the survival rate curve of MG-63 cells after PDT was drawn.Cell morphological changes were observed by microscope.The OD492 values at early time after PDT were measured by MTT assay.Results The survival rate of cells after PDT was 48.09% and significantly decreased when the concentration of PSD-007 was 2 μg/ml and the light energy was 6 J/cm2.Also,cellular morphology changed significantly after PDT.The apoptosis at the early time after PDT was not obivious.Conclusions PSD-007 could obviously inhibit the growth of human osteosarcoma cell line MG-63 in vitro.
