Correlation analysis of PIK3CA protein positive expression rate with pathological characteristics and anti-tumor immune response in cervical cancer

Abstract:Objective To study the correlation of phosphatidylinositol 3-kinase catalytic alpha polypeptide(PIK3CA)protein positive expression rate with pathological characteristics and anti-tumor immune response in cervical cancer.Methods A total of 108 patients with cervical cancer who underwent radical surgery in the Second Affiliated Hospital of Hainan Medical University from January 2017 to March 2018 were retrospectively selected as the cervical cancer group and cervical cancer tissues were collected,68 patients who underwent surgical resection due to uterine fibroids at the same time were selected as the control group and normal cervical tissues were selected.The positive expression rate of PIK3CA protein in cervical cancer tissues and normal cervical tissues was detected by immunohistochemical staining,the serum contents of immune cytokines interleukin(IL)-2,IL-4,IL-5,IL-10,tumor necrosis factor-α(TNF-α),Interferon-γ(IFN-γ)were detected by ELISA.The pathological characteristics of patients with cervical cancer were collected,disease-free survival and overall survival of patients with cervical cancer were followed-up.Results The positive expression rate of PIK3CA protein in cervical cancer tissue was 77.78%,which was higher than that in normal cervical tissue(50.00%),and the difference was statistically significant(P<0.05).The proportion of FIGO stage IIA,vascular invasion,and lymph node metastasis in cervical cancer tissue positive patients with PIK3CA protein were 78.57%,48.81%,and 40.48%,which were higher than those in PIK3CA protein-negative patients(54.17%,20.83%,16.67%),the differences were statistically significant(P<0.05),and there was no significant difference in pathological type,differentiation degree and PIK3CA protein-negative patients(P>0.05).The serum levels of IL-2,TNF-αand IFN-γin patients with positive PIK3CA protein in cervical cancer tissue were(3.04±0.89)pg/mL,(10.61±1.83)ng/mL,(13.24±2.92)ng/mL,which were lower than those in patients with negative PIK3CA protein[(4.13±1.14)pg/mL,(13.84±4.52)ng/mL,(17.02±4.95)ng/mL],serum IL-4,IL-5,IL-10 levels were(2.98±0.52),(4.42±1.09),(6.76±1.14)pg/mL,which were higher than those in patients with negative expression of PIK3CA protein[(2.24±0.77),(3.18±0.71),(5.14±0.93)pg/mL],the differences were statistically significant(P<0.05).The cumulative disease-free survival rate and cumulative overall survival rate of patients with positive PIK3CA protein in cervical cancer tissue during follow-up were lower than those of patients with negative expression of PIK3CA protein,and the difference was statistically significant(P<0.05).Conclusion The increased expression of PIK3CA in cervical cancer relates with pathological progress,decreased anti-tumor immune response and decreased long-term survival rate.
