摘    要:目的探索大鼠胚胎神经干细胞的培养、传代和冻存、复苏的方法,以及观察胚胎神经干细胞在自然条件下分化为神经元细胞和神经胶质细胞的过程.方法从胚胎大鼠脑组织中分离得到神经干细胞,采用无血清培养技术进行培养、传代和鉴定.诱导分化后采用SABC法对分化的细胞进行神经元特异性烯醇化酶(NSE)、神经胶质酸性蛋白(GFAP)检测以作细胞鉴定.结果成功培养出大鼠胚胎神经干细胞,并对其进行传代、冻存及复苏,培养的细胞能分化为神经元细胞和神经胶质细胞.结论大鼠胚胎神经干细胞能在体外适宜的培养条件下进行长期的培养、传代及冻存、复苏,并具有多向分化潜能.

Abstract:Objective To observe the growth and differentiation of neural stem cells originated from rat embryo brains. Methods The cells isolated from rat embryo brains were incubated with serum free medium. The capacity of proliferation and the characteristics of differentiation of the neural stem cells were examined with immunocytochemistry. Results We incubated the neural stem cells, the cells were able to proliferate and be cryopreserved and resuscitated. The differentiated cells expressed NSE and GFAP, characteristic for neurons and glial cells respectively. Conclusion The neural stem cells derived from rat embryo brains in this study are able to proliferate and multipotential for differentiation.
