摘    要:离心沉降分析技术是分子生物学基本实验技术之一。3年来,我们用Model-282分析型超速离心机测定了蛋白质、核酸及多糖的分子量(M,沉降-扩散法)和沉降系数(S)80多次,均获得满意结果,为鉴定这些生物大分子的理化特性提供基本数据,本文对该技术原理、方法及测定实例作详细介绍。实验表明,它是测定生物大分子特性的简便、精确、可靠的基准方法。

Abstract:During the past three years, we measured the moleoular weight(M.W) and sedimentation coefficient (S) of some proteins, nucleic acids and polysacchari des up to 80 times, using Hitachi Model-282 analytical ultracentrifuge. In this paper, we introduced principle and method in detail for the determination of sedimentation coefficient by Schlieren and Absorption scanning optical systems and for the determination of molecular weight by the method of combining boundary sedimentation with synthetic boundary procedure. All of the results obtained are satisfactory. Experimental error amounts to 3-5% permitted by this instrument. We expect that the precision of results may be improved by changing our tools used for measurement of the related parameters.
