生殖器疱疹患者外周血淋巴细胞B7-CD28/CTLA-4的检测分析

摘    要:目的研究外周血B7-CD28/CTLA-4(CD152)T淋巴细胞活化协同刺激分子在生殖器疱疹(Genital herpes,GH)患者发病机制中的作用。方法用流式细胞仪检测51例GH患者和15例健康体检者外周血淋巴细胞CD80/86及CD4+/CD8+T淋巴细胞中B7-CD28/CTLA-4的表达。结果GH患者恢复组与对照组比较,CD80、CD86的阳性表达较对照组明显升高,差异有统计学意义(P〈0.05)。其余各组间CD80、CD86比较差异无统计学意义。恢复组CD8+/CD28阳性表达显著高于非复发组(P〈0.05);非复发组显著高于对照组(P〈0.05);复发组的CD4+/CD152、CD8+/CD152的阳性表达显著高于非复发组(P〈0.05)。其余各组间CD4+/CD152、CD8+/CD152及CD4+/CD28、CD8+/CD28表达差异无统计学意义。结论单纯疱疹病毒感染可诱导CD80、CD86、CD28和CD152活化增殖,高表达负性共刺激分子和分泌抑制性细胞因子,通过多种机制抑制机体的抗病毒免疫应答。

Abstract:Objective To investigate the role of B7-CD28/CTLA-4(CD152),co-stimulatory molecules of T lymphocytes in the pathagenesis of genital herpes(GH).Methods Flow cytometry was utilized to detect the expression levels of CD80/86 in peripheral blood lymphocytes and of CD28/CD152 in CD4+/CD8+T lymphocytes from 51 patients with GH and 15 healthy volunteers as controls.Results The frequencies of CD80,CD86 lymphocytes were significantly higher in the recovery group than those in the control group(P〈0.05).No significant difference was found for the frequencies of CD80,CD86 lymphocytes between the recurrent or non-recurrent group and the control group.The frequencies of CD8+/CD28 cells were significantly higher in the recovery group than those in the non-recurrent group(P〈0.05).The frequencies of CD8+/CD28 cells were significantly higher in the non-recurrent group than those in the control group(P〈0.05).The frequencies of CD4+/CD152,CD8+/CD152 cells were significantly higher in the recurrent group than those in the non-recurrent group(P〈0.05).No significant difference of those cells was found between the recurrent and the control group.Conclusion Herpes simplex virus infection may result in the activation and proliteration of CD80,CD86,CD28 and CD152,expression of high level negative costimulatory molecules and secretion of suppressive cytokines,which may lead to the inhibition of antiviral immune responses via multiple mechanisms.
