男性不育患者精子XY染色体的荧光原位杂交分析

摘    要:目的探讨用荧光原位杂交分析的方法来检测少、弱、畸形精子症患者中精子XY染色体畸变率。方法将30例少、弱、畸形精子症患者和20例正常生育男性精液精子标本用荧光原位杂交分析的方法检测精子X和Y染色体的数目畸变的发生率,并将结果进行分析、比较。结果研究组单倍体精子数32 189个,其中单倍体X精子数占(45.34±6.56)%,单倍体Y精子数占(47.32±7.18)%;二倍体精子率为(0.168±0.059)%,精子X和Y染色体数目畸变率为7.34%。对照组单倍体精子数为19 246个,其中单倍体X精子数为(46.87±5.54)%,单倍体Y精子数为(49.32±6.17)%;二倍体精子率为(0.074±0.038)%,精子X和Y染色体数目畸变率为3.81%。两组二倍体精子率、精子数目畸变率差异有统计学意义(P〈0.05)。结论少、弱、畸形精子症患者中精子X和Y染色体数目畸变率比正常生育人群高。

Fluorescence in situ hybridization of sperm chromosome X and Y in male infertility of oligoasthenoteratozoospermia (OAT)

Abstract:Objective To detect the rates of numerical chromosome aberration of sperm chromosome X and Y in oligoasthenoteratozoospermia(OAT) patients with fluorescence in situ hybridization(FISH).Methods Thirty OAT and 20 normal fertility men’s sperm samples were collected and treated with FISH,then the rates of numerical chromosome aberration of sperm chromosome X and Y.was detected.Results There were 32 189 haploid sperms were counted in this experiment,the haploid sperm X chromosome rate was(45.34±6.56)%,Y chromosome rate was(47.32±7.18)%,diplont sperm rate was(0.168±0.059)%,the sperm numerical aberration rate was 7.34%.In control group,19246 haploid sperms were counted and the haploid sperm X chromosome rate was(46.87±5.54)%,Y chromosome rate was(49.32±6.17)%,diplont sperm rate was(0.074±0.038)%,the sperm numerical aberration rate was 3.81%.There were significant differences in the diplont sperm rate and sperm numerical aberration rate between two groups(P〈0.05).Conclusion The rate of numerical chromosome aberration of sperm chromosome X and Y in OAT is higher than that of normal fertility men.
