妊高症患者及新生儿脐血VE-cadherin和HMGB1的检测

目的分析妊高症患者及新生儿脐血血清血管内皮细胞钙黏蛋白（VE-cadherin）和高迁移率蛋白-1（HMGB1）的变化。方法采用酶联免疫吸附法（ELISA）测定98例妊高症患者（包括妊娠期高血压40例、轻度子痫前期30例和重度子痫前期28例）及30例正常对照组产妇及新生儿脐血血清VE-cadherin和HMGB1含量。结果VE-cad herin和HMGB1在妊高症组及正常对照组及新生儿脐血血清中均有表达,VE-cadherin在三组妊高症病患者血清中含量与正常对照组比较,差异有显著意义（t=5.83,8.41,9.04,P均〈0.05）。随着病情越严重,VE-cadherin水平越高,三组之间差异有统计学意义（t=6.23,8.56,P均〈0.05）;HMGB1与VE-cadherin分布高度一致,并呈正相关（r=0.59,P〈0.01）。VE-cadherin和HMGB1在妊高症各组新生儿脐血中的含量,差异无统计学意义（P〉0.05）。结论VE-cadherin和HMGB1的高表达,显示妊高症病患者内皮细胞损伤,是妊高症内皮损伤炎症标记物,为妊高症的治疗和预防开辟一条新途径

Expression of VE-cadherin and HMGB1 in peripheral blood and umbilical cord blood of pregnancy-induced hypertension patients

Objective To investigate the expression and significance of VE-cadherin and high mobility group box-1（HMGB1） in intravenous serum and umbilical cord blood of patients with pregnancy-induced hypertension syndrome,PIH.Methods Contents of VE-cadherin and HMGB1 in intravenous serum and umbilical cord blood from 98 pregnancy-induce hypertension patients which were divided into gestational hypertension40,mild and severe preeclampsia groups 30 and 28 patients were detected by enzyme-linked immunosorbent assay（ELISA） respectively.30 normal pregnant women were taken as control group.Results VE-cadherin and HMGB1 expressed in both patients and controls.Serum contents of VE-cadherin in patients and controls had significant difference by comparison（t=5.83,8.41,9.04,all P〈0.05）.Moreover,there were differences among the contents of three groups（t=6.23,8.56,all P〈0.05）.Contents of VE-cadherin and HMGB1 of patients with hypertensive disorders in pregnancy showed a positive correlation（r=0.59,P〈0.01）.There were no significant differences in contents of VE-cadherin and HMGB1 in umbilical cord blood of patients with hypertensive disorders in pregnancy in three groups（P〉0.05）.Conclusion The high expression of VE-cadherin and HMGB1 in pregnancy-induce hypertension patients indicates the injury of vascular endothelium cells and VE-cadherin and HMGB1 play a role in the process of injury.
