三氧化二砷对人肾母细胞瘤细胞作用及机制的研究

目的研究三氧化二砷（arsenictrioxide，As203）对体外人肾母细胞瘤（Nephroblastoma／Wilms’tumor，WT）SK—NEP-1细胞作用及CyclinDl表达的影响。方法采用不同浓度的三氧化二砷作用体外培养人肾母细胞瘤WT细胞，应用MTT比色法检测细胞增殖的影响；RT-PCR法检测CyclinDlmRNA的表达。结果MTT检测对照组及不同药物浓度组增殖抑制率分别为（10．22±0．70）％、（20．76±3．46）％、（31．09±2．95）％、（40．04±2．30）％、（53．04±3．02）％、（64．61±0．95）％；RT-PCR法检测CyclinDlmRNA的表达，对照组及不同药物浓度组的灰度值分别为：1．25±0．07、1．03±0．08、0．90±0．09、0．73±0．08、0．48±0．09、0．26±0．07。以上检测各组间比较差异均有统计学意义（P〈0．05）。结论三氧化二砷可明显抑制WT细胞增殖，其机制可能与CyclinD1表达水平降低有关。

Effects of Arsenic Trioxide on Cells and Expressions of CyclinD1 in Human Nephroblastoma WT Cells

Objective To investigate the effects of arsenic trioxide on cells and expressions of CyclinD1 in human Nephroblastoma WT cells. Methods WT cells were exposed to various concentrations of arsenic trioxide for certain time. MTT colrimetric assay was performed for the proliferation inhibition rates. RT-PCR was used to test the expression of CyelinD1 mRNA in WT cells. ResuLts The proliferation inhibition rates of control group and different concentrations of arsenic trioxide groups were （10.22±0.7）%,（20.76±3.46）%,（31.09±2.95）%,（40.04＋2.30）%,（53.04±3.02）%,（64.61±0.95）%; To the expression of CyelinD1 mRNA,their grey levels were： 1.25±0.07,1.03±0.08,0.90±0.09,0.73±0.08,0.48±0.09,0.26±0.07.To all the resμLts,MlaLtipe comparison had significant differences（P〈0.05）. Coneluslons Arsenic trioxide can inhibit WT cells proliferation. Its molecμLar mechanisms might be related to decrease the expression of CyclinD 1 mRNA.
