促红细胞生成素对戊四氮致癫痫幼鼠脑保护作用研究

目的]探讨不同时间点给予促红细胞生成素(EPO)对戊四氮(PTZ)致癫幼鼠癫痫(EP)后的脑保护作用。方法]60只SD幼鼠随机分为6组,实验组(A～E组)注射PTZ50mg/(kg.次),对照组(F组)注射等量生理盐水。实验组分为单纯致癫组(A组)及EPO干预组(B～E组),于癫痫发作前24h(B组)、发作后0h(C组)、6h(D组)、9h(E组),分别注射rhEPO5000u/(kg.次);均于EP后24h取海马,用酶联免疫吸附法检测神经元特异性烯醇酶(NSE)和S100β水平。结果]B组癫痫发作程度及严重性较A组减轻。海马NSE及S100β浓度A、E组均高于B、C、D和F组,而B、C、D组与F组间类似。结论]EPO可能具有抗癫痫作用,对PTZ致痫幼鼠具神经保护作用,尤以EP发生后6h内给药作用最佳。

Study on Neuro-protection Effects of Erythropoietin in Rats with Epilepsy Induced by Pentylenetetrazo

Objective] To evaluate neuro-protection effects of erythropoietin（EPO）at different time points before or after epilepsy（EP）seizure.Methods] Totally 60 male SD rats were divided randomly into 6 groups：experimental groups（A-E）were injected with pentylenetetrazol（PTZ）50 mg/kg,and the control group（F）was injected with normal saline.The experimental groups were subdivided into epileptic group（A）and intervention groups（B-E）which were injected with rhEPO 5 000 u/kg,24 h before seizure（B）,and 0 h（C）,6 h（D）and 9 h（E）after seizure.Rats were sacrificed 24 h after seizure,the hippocampus were resected for measuring the levels of neuron-specific enolase（NSE）and S100βprotein using ELISA.Results] The level and severity of seizure in group B were significantly lighter than that in group A;The levels of NSE and protein S100β in group A and E were significantly higher than those in group B,C,D and F,and no significant differences among group B,C,D and F.Conclusion] EPO might have anti-EP and neuro-protection effects,and the better effects were within 6 h after seizure.
