蓖麻毒素及其A、B链的提取分离纯化的研究

摘    要:本文研究蓖麻毒素及其A、B链的提取、分离、纯化,根据B链能与Sepharose-6 B上的半乳糖结合,而A链不具有这一性质,用β-巯基乙醇使蓖麻毒素直接在Sepharose-6B柱上断链,然后分别用Tris-HCl和Tris-HCl-半乳糖溶液洗脱,首次把蓖麻毒素的制备、纯化以及A、B链的断链、分离结合起来,大大简化了工艺。

PURIFICATION OF RICIN, RICIN A AND B CHAINS

Abstract:This paper describes a method for the preparation of ricin and its constituent peptide chains, namely ricin A and B chains. The purification involved extraction from castor beans by using 5% acetic acid solution and chromatography on a CM52 column and subsequently on a sepharose-6B column. The sepharose-6B column was first washed with 10 mmol/L Tris-HC1 (pH=7.7), then the intact ricin was split into A and B chains on the sepharose-6B column with 2-mercaptoethanol. The A chain was eluted with Tris-HCl buffer containing 2-mercaptoethanol. The B chain was eluted with Tris-HCl buffer containing galactose. The B chain was further purified by chromatography on a DEAE-csllulose column. The A chain was further purified by chromatography on a DEAE-cellulose column and a CM 52 column.
