乙酰紫堇灵对小鼠对乙酰氨基酚肝中毒的保护作用

摘    要:乙酰紫堇灵(Ace)是从中药紫堇中提取的生物碱,本文研究了Ace对小鼠对乙酰氨基酚(APAP)肝损伤的影响。Ace显著降低APAP引起的小鼠血清谷丙转氨酶升高,病理观察肝细胞损伤明显减轻。Ace显著防止小鼠ipAPAP引起的肝脏谷胱甘肽耗竭,降低小鼠血中APAP浓度。Ace对肝脏细胞色素P450总量及谷胱甘肽巯基转移酶(GSH-S-T)有显著诱导作用,而对代谢APAP的P450同工酶ⅡE1有抑制作用。以上结果表明Ace对小鼠APAP中毒的保护作用与其选择性地调节P450同工酶,诱导GSH-S-T,从而加快APAP在体内的解毒代谢密切相关。

Abstract:Acetylcorynoline (Ace)isolated fromCorydalis bulleyana Diels is an alkaloid.The effects ofAce on liver injury induced by paracetamol fAPAP)were studied in mice.The elevated serum glutamicpyruvic transaminase level induced by APAP wassignificantly decreased by Ace pretreatment.Pathologi-cal damages of the hepatocytes were also lessened.Thedepletion of liver glutathione induced by APAP wasprevented.Blood APAP concentration of Acetreatedmice was much lower than that of control.Ace treat-ment induced significant increase of liver glutathioneStransferase(GSHST)and microsomalcytochrome P450,while P450 isoenzyme Ⅱ E,activitywas slightly inhibited.These results suggest that theprotective action of Ace against APAP hepatotoxicitymay attribute to its selective regulation on livercytochrome P450 isoenzymes and induction ofGSHST.
