Effect of nimodipine on pial arteriolar responseto endothelin1-induced vasospasm

It was studied in anesthetized rats with a cranial window for the observation of the pialmicrocirculation of the parietal cortex。The cranial windowwas suffused with endothelin一1(ET一1).A low coneentra-tion of ET-1(0.1 nmol·L-1)produced modest(8.9±s 3.4%) dilatation of pial arterioles。Higher concentrationsof ET一1(1,10 and 100 nmol·L-1) constricted pialarterioles with decrease in diameter of 28±s4%,47±s 5%and 78±s5%,respectively. ET一1 Was found to be less active at constricting the rat pial veins on the cerebrum。Intravenous administration of nimodipine (0.3mg·kg-1 ) attenuated,but did not completely abolish,the constrictionindueed by ET-1(100nmol·L-1)。
