卵巢癌HER-2/neu表达与肿瘤免疫细胞分布关系

摘    要:目的 研究上皮性卵巢癌细胞原癌基因HER-2/neu表达与肿瘤微环境免疫活性细胞分布的关系,探讨过度表达原癌基因HER-2/neu的上皮性卵巢癌临床分期晚、预后差的可能原因。了解过度表达HER-2/neu基因的上皮性卵巢癌肿瘤微环境的免疫细胞分布。方法 用流式细胞仪检测上皮性卵巢癌细胞原癌基因HER-2/neu的表达,同时用免疫组化方法检测肿瘤微环境中LCA^ 、CD^ 3细胞分布情况。结果 30%卵巢癌细胞原癌基因HER-2/neu呈过度表达,且卵巢癌细胞原癌基因HER-2/neu表达与肿瘤微环境中LCA^ 、CD^ 3细胞分布呈负相关,前者r=-0.630,P=0.0001,后者r=-0.552,P=0.0009。结论 研究结果提示过度表达原癌基因HER-2/neu的上皮性卵巢癌患者肿瘤微环境中浸润免疫细胞数减少,两者存在着一定的内在联系,可能发生肿瘤细胞的免疫逃逸。

Abstract:Purpose To determine the correlation between expression of proto-oncogene HER-2/neu on ovarian tumor cells and T-cell infiltration in vivo ovarian tumor micro-environment. Methods Thirty three samples of primary epithelial ovarian can cers were analysed with flow cytometry for proto-oncogene HER-2/neu expression.Immunohistochemical analysis was performed using monoclonal antibodies that recognize leukocyte differentiation antigens (LCA,CD 3) on cryostat sections of prinmary epithelial ovarian cancinoma.The results were compared with the expression of proto-oncogene HER-2/neu expression.which provises the statue of micro-environment in ovarian carcinoma patients which have overexpression of proto-oncogene HER-2/neu. Results Close correlation were revealed between the expression of proto-oncogene HER-2/neu and the numbers of infiltrating LCA positive cells (r=-0.680,P=0.000 1) and CD+ 3 cells (r=-0.552 39,P=0.000 1). Conclusions The study revealed that expression of proto-oncogene HER-2/neu has some inherent correlation with the status of human immunity in tumor micro-environment on ovarian carcinoma.
