中药黄芩HPLC指纹图谱的化学轮廓及其影响因素的研究

摘    要:目的建立中药黄芩HPLC指纹图谱方法,研究指纹图谱化学轮廓及其影响因素,为黄芩HPLC指纹图谱方法的推广应用提供参考依据。方法采用HPLC指纹图谱分析方法对7个省区83份黄芩(Scutellariabaicalensis根)的化学成分进行测定,考察色谱柱、色谱仪、洗脱梯度、检测波长等因素对指纹图谱的影响,对色谱方法的普适性和耐用性作出系统评价。结果用83份黄芩样品建立的黄芩HPLC指纹图谱中有29个共有峰。利用对照品和LC—MS技术对黄芩指纹谱中的27个化合物进行了指认和鉴定;在一定的条件下所建立的指纹谱方法具有良好的精密度、重复性.黄芩供试品溶液在72h内稳定性良好。在考察各因素中发现,检测波长对指纹谱的化学轮廓影响很大;分别变换色谱仪、色谱柱或洗脱梯度等色谱条件对指纹谱化学轮廓产生的影响亦较大。结论所建立的黄芩HPLC指纹图谱方法在一定条件下具有很好的稳定性和实用性,可用于黄芩药材的化学轮廓研究,进而评价黄芩的质量。但是指纹图谱在实际应用中须严格控制规定的测定条件,以确保所得指纹图谱稳定可靠。

Abstract:Objective To establish HPLC fingerprint approach for studying the chemical profile and influencing factors of Seutellaria baicalensis and provide a reference method for the application of the HPLC fingerprint. Methods The chemical compositions of 83 samples, which were collected in seven provinces, were analyzed using HPLC fingerprint method. Several influencing factors including chromatographic column, HPLC instrument, elution gradient and detec- tion wavelength were investigated. Universality and durability of the method were systematically evaluated. Results 29 common peaks were found in the chromatographic fingerprints of 83 samples and 27 compounds were identified by ref- erence substances and LC-MS. The fingerprint method had a good precision and reproducibility, and the samples were stable in 72 hours. The chemical profile of the fingerprints were greatly influenced by the detection wavelength. There were great differences in the fingerprints when the chromatographic conditions were changed, such as chromatographic column and elution gradient. Conclusion The HPLC fingerprint method established has a good stability and practical- ity, which can be used to evaluate the quality of Scutellaria baicalensis. To ensure the stability and reliability of the fingerprint, the required determination conditions of the fingerprint should be well controlled in real application.
