摘    要:小鼠ig 3-乙酰乌头碱的LD_(50)为3.09mg/kg,其他各种注射途径的LD_(50)在0.40-0.70mg/kg间,约为ig的13-23%;大鼠LD_(50)为小鼠的53-74%。在亚急性毒性实验中,3-乙酰乌头碱中毒剂量可致大鼠呼吸抑制而死亡,家兔除有呼吸抑制外尚出现ECG的异常。组织病理学变化主要为心肌细胞变性,肝细胞轻度损伤。阿托品加人工呼吸对3-乙酰乌头碱所致的大鼠和兔的呼吸抑制有较好的对抗效果。致畸实验未见胎仔畸形。

Abstract:3-Acetylaconitine(AAC) is a new alkaloid isolated from Aconitum flavum. Its acute ig LD50 was 3.09 mg/kg in mice, while the LD50 of the drug administered by other routes were 13-23 percent of ig LD50. In subacute experiments, the main toxic manifestations of AAC were respiratory depression which usually caused death in rats and abnormal ECG in rabbits. In histo-pathological examinations all of the animals treated with AAC showed degenerative change in heart muscle cells and slight damage in livercells. It has been found that atropine plus artificial respiration were effective in increasing survival number of intoxicated rabbits. No teratogenic effect of AAC was observed on mice or rabbits. In the positive control with cod-liver oil extract a lot of external malformations and cleft palate in the fetuses of mous had been found.
