摘    要:目的应用荧光定量PCR技术分析MicroRNA-29b(miR-29b)在Apo-E基因敲除小鼠的动脉粥样硬化(atheroscle—rosis)模型斑块血管中的表达情况,探讨miR-29b在动脉粥样硬化发生、发展过程中的作用。方法8只5周龄、雄性SPF级载脂蛋白E基因敲除小鼠(C57BL/6J—ApoE-/-,apolipoproteinE—deficientmice)为实验组(FS组)。8只雄性、SPF级、相同周龄的C57BL/6J小鼠为对照组(NS组)。高脂(1%胆固醇+15%猪油)喂食15、20周龄取材检测。结果FS组和NS组体重(29.10±2.53)g和(33.20±5.18)羽无明显差异(P〉0.05);FS组较NS组血清总胆固醇((13.52±1.58)mmol/L和(2.12±0.35)mmol/L]显著增高(P〈0.01);FS组主动脉出现典型的无动脉粥样硬化斑块;NS组主动脉管壁正常,无动脉粥样硬化斑块;FS组主动脉弓miR-29b的表达较NS组平均吸光度值:(2.10±0.27)和(17.56±1.07)1明显降低(P〈0.01)。结论在高脂喂食的载脂蛋白E基因敲除小鼠动脉粥样硬化模型中.miR-29低表达可能与动脉粥样硬化斑块的形成有关。

Abstract:Objective To determine the role and relationship of MicroRNA-29b (miR-29b) in the development process with the atherosclerotic were used fluorescence quantitative PCR technical in the expression of miR-29b on atherosclerotic aorta in Apolipoprotein E-deficient mice. Methods 8 five-week-old male mice which were knocked out apo-E gene grade of SPF C57BL/6J were grouped as the experiment group (FS group). 8 male mlce of the same age grade of SPF C57BL/6J were grouped as the control group (NS group). Two groups of mice were fed up with high fat diet for fifteen weeks (1%cholesterol+15%lard). When the mice were twenty weeks, took some samples and did the test. Results There was no significant difference between FS group and NS group in the body weight (29.10±2.53) g, (33.20±5.18)g](P 〉 0.05). Serum total cholesterol was significantly higher in the FS group than that in the NS group (13.52±1.58)mmol/L, (2.12±0.35) mmol/L] (P 〈 0.01). There were typical atherosclerotic plaque in aorta in the FS group, but they were normal and there were no atherosclerotic plaque in the NS group. The expression of miR-29b in aortic arch was significantly lower in the FS group than that in the NS group IOD: (2.10±0.27), (17.56±1.07)] (P 〈 0.01). Conclusion The forming of atherosclerotie plaque in atherosclerosis model which Apolipoprotein E-deficient mice were fed with high fat diet concern to the lower expression of miR-29b.
