香菇多糖对巨噬细胞一氧化氮生成和细胞内还原型谷胱甘肽的影响

摘    要:目的 为探讨香菇多糖(lentinan,LTN)激活小鼠腹腔巨噬细胞一氧化氮(NO)生成的作用机制。方法 观察LTN对小鼠腹腔巨噬细胞NO生成与细胞内还原型谷胱甘肽(GSH)的影响及其关系。结果 ⑴LTN对小鼠腹腔巨噬细胞NO生成具有明显的促进作用,同时细胞内GSH浓度随NO生成增加而减少;⑵NO生成抑制剂能抑制LTN刺激的小鼠腹腔巨噬细胞NO生成和细胞内GSH水平降低;⑶GSH去除剂能抑制巨噬细胞NO生成。结论 研究表明LTN促进小鼠腹腔巨噬细胞NO生成并同时消耗细胞内GSH,细胞内GSH水平可能与NO的生成有密切关系。提示细胞内GSH可能起到调节巨噬细胞NO生成和保护宿主细胞免受NO介导的细胞毒作用。

Abstract:Object To investig ate the mechanisms of action of lentinan(LTN)activated mouse peritoneal macrophagesto produce nitricoxide(NO).Methods The effects of LTN on NO out put and intrace llular glutathione(GSH)in mouse peritone almacrophages and the correlation of them were studied. Results (1)LTN can significantly in crease production of NO in mouse peritoneal macrophages,decre as elevel of in tracellular GS H followed incre ase of NO production.(2)The above effect can be blocked efficiently by the NO production inhibitors.(3)NO production can be inhibited by G S H-lowering drugs.Conclusion LTN increase NO production with depletion of intracellular G S H in the activated mouse peritoneal macrophages. I tsuggested that in trace llular GSH plays an importantrole in regulation of NO production and protection of host cells from cytotoxic at tackinduced by NO.
