| 摘 要： | 目的探讨阿米洛利(Amiloride)与川芎嗪(Ligustrazine)联用对大鼠肝星状细胞(hepatic stellate cells,HSC)氧应激所致纤维化的作用。方法使用FeNTA建立HSC氧应激模型。检测HSC的增殖、Ⅰ型胶原、丙二醛(MDA)水平、超氧化物歧化酶(SOD)活力;药物对HSC的毒性作用。结果阿米洛利和川芎嗪可以明显抑制HSC增殖,可明显降低MDA和Ⅰ型胶原的水平和提高SOD活力,且合用组优于单用组。结论作用于HSC不同靶点的抗纤维化药物合用优于单用。 |

| Abstract: | Objective To investigate the effects of ligustrazine and amiloride on the fibrogenic effect of oxidative stress in rat hepatic stellate cells(HSC) in vitro.Methods Oxidative stress model was established by incubating HSC with ferric nitrilotriacetate complex(FeNTA).MTT colorimetry was used for assaying proliferation of HSC.Cytotoxicity was measured by LDH colorimetry.Collagen typeⅠaccummulation in the culture media was measured by ELISA.MDA level and SOD activity in the culture media were measured by their reagent boxes.Results Ligustrazine and amiloride significantly inhibited the level of collagen typeⅠand MDA and enhanced the activity of SOD.Combination group had better effects on the fibrogenic effect of oxidative stress in rat HSC than above each one.Conclusion The effects of combination of antifibrosis drug on different targets of HSC are superior to those of each one. |
