归术活血胶囊对脑缺血再灌注损伤模型大鼠的保护作用

目的:探讨归术活血胶囊对大鼠脑缺血再灌注损伤的保护作用。方法:建立大鼠大脑动脉栓塞模型,随机将大鼠分为模型组、尼莫地平阳性对照组及归术活血胶囊高、中、低剂量组,另设假手术组。各组灌胃给予水或相应药物,将皮层及海马组织分为损伤侧与对照侧,测定两侧天冬氨酸(Asp)、谷氨酸(Glu)、丙二醛(MDA)、乳酸脱氢酶(LD)、肿瘤坏死因子α(TNF-α)和谷胱甘肽过氧化物酶(GSH-Px)的含量或活性,并以红四氮唑(TTC)染色法测定脑梗死面积,以烘干法测定脑含水量。结果:与模型组比较,归术活血胶囊组(其中一组或几组)皮层Asp含量无明显变化,而海马组织损伤侧Glu含量明显降低(P<0·05),皮层损伤侧TNF-α含量明显增加(P<0·05),皮层及海马组织两侧MDA含量和GSH-Px活性显著降低(P<0·05),皮层损伤侧LD含量轻微升高,对照侧LD含量明显下降(P<0·05),皮层两侧K+-Na+-ATP酶活性降低(P<0·05),脑含水量降低且脑梗死面积明显减小(P<0·05)。结论:归术活血胶囊对模型大鼠局灶性脑缺血再灌注损伤具有保护作用。

The Protective Effect of Guishu Blood - activating Capsules on Cerebral Ischemical Reperfusion Injury Model Rats

OBJECTIVE:To investigate the protective effect of Guishu blood-activating capsule on cerebral ischemical reperfusion injury of rats.METHODS:The cerebral embolism model rats were established,the rats were randomly divided into model group,nimodipine positive control group,Guishu blood-activating capsule(high,medium and low dosage groups,reˉspectively)group and with another sham operation group established.Each group was administered intragastrically with water or the corresponding drugs,the cortexes and hippocampus tissues of the rats were divided into the injured side and the control side,the contents of aspartate(Asp),glutamate(Glu),malonaldehyde(MDA),lactate dehydrogenase(LD),the tumor necrosis factorα(TNF-α)and the activity of glutathione peroxidase(GSH-Px)were determined,the cerebral infarct size was determined by red tetrazoline staining method and the cerebral water content was determined by oven dry process.REˉSULTS:Compared with the model group,in one or several sub-groups of Guishu group,no marked change was noted in the content of Asp while that of the Glu in the injured side of the hippocampus tissues decreased significantly(P<0.05),the conˉtents of TNF-αincreased markedly in the injured side of the cortex(P<0.05),the content of MDA and the GSH-Px activity in both sides of cortexes and hippocampus tissues deceased markedly(P<0.05),the content of LD in the injured side of the cortex increased slightly while that in its control side decreased significantly(P<0.05),the activities of K + -Na + -ATP deˉcreased in both sides of cortexes(P<0.05),the cerebral water contents decreased and the cerebral infarct size diminished markedly(P<0.05).CONCLUSION:Guishu blood-activating capsule has protective effect on focal cerebral ischemical reperfusion injury model rats.
