补肾醒脑解毒方对大鼠单纯疱疹病毒脑炎脑内NGF、BDNF表达的影响

摘    要:目的:观察补肾醒脑解毒方对大鼠单疱病毒脑炎后脑内神经营养分子如NGF、BDNF表达的调节作用。方法:以HSV-1悬液经角膜划痕接种结膜囊内复制大鼠HSV脑炎模型,用Northern杂交与ELISA方法检测实验动物治疗前后脑内NGF、BDNF的mRNA与蛋白含量。结果:HSV脑炎大鼠及正常大鼠服补肾醒脑解毒方后脑内NGF、BDNF mRNA及蛋白均增高。结论:补肾醒脑解毒方对HSV脑炎后大鼠脑内NGF、BDNF表达有促进作用,提示补肾醒脑解毒方对HSV脑炎后遗症潜在着良好治疗作用。

Abstract:Objective: Investigating the effects of Bushen Xingnao Jiedu Fang on expressions of NGF and BDNF in rat brain after infection of herpes simplex virus type 1 (HSV 1). Methods: Rats with 20 day age were anesthetized intravenously with pentobarbital sodium and their right conjunctival sac was infected with 50ul of virus suspension (containing about 10 4 TCID 50 HSV 1 viron) after the cornea scarified.The mRNAs of NGF and BDNF were detected with Northern hybridization, and the protein of NGF and BDNF were detected with ELISA. Results: (1) The expression of NGF and BDNF was decreased at the day 20 after HSV 1 infection by the cornea scarified. (2) The expression level of NGF mRNA and BDNF mRNA and the protein of NGF and BDNF were increased in brains of both HSV 1 infected rats and normal rats after 30 days of treatment with the decoction of Bushen Xingnao Jiedu Fang. Conclusion: Bushen Xingnao Jiedu Fang was found to have the effects of up regulating expressions of NGF and BDNF in rat brain after HSV 1 encephalitis. The results suggested that Bushen Xingnao Jiedu Fang was a potential therapy for neurological deficiency after HSV encephalitis.
