牛珀至宝微丸对内毒素休克鼠肺组织中一氧化氮合酶活性的影响

摘    要:目的:观察牛珀至宝微丸(由水牛角、麝香、地黄、牛砂、牛黄、郁金等组成)对内毒素休克时肺损伤的保护作用和肺一氧化氮合酶(NOS)表达的影响。方法:静脉注射脂多糖(LPS)1.5mg/kg、腹腔注射D-氨基半乳糖(D-Gal)100mg/kg造成内毒素休克模型,用牛珀至宝微丸和氨基胍(AG)作对照干预处理,用硝酸还原酶法检测血浆一氧化氮(NO)浓度,同位素标记法测肺组织匀浆NOS活性,免疫组化方法检测诱生型一氧化氮合酶(iNOS)、内皮型一氧化氮合酶(eNOS)在肺组织内的表达。结果:牛珀至宝微丸可以降低内毒素休克时血浆NO浓度,降低肺组织iNOS活性,使肺内iNOS表达减弱、eNOS表达增强,肺损伤减轻。结论:牛珀至宝微丸可以减轻内毒素休克造成的肺损伤,这种保护作用可能是通过调节肺组织NOS表达而产生的。

Abstract:Objective] To observe the effect of Niupo Zhibao Pellet (NZP) , mainly composed of Cornu Bubali, Mos-chus, Radix Rehmanniae, Cinnabaris and Radix Curcumae, on lung injury and the expression of NO synthetase (NOS) in lung tissue of rats with endotoxic shock. Methods] Rats were randomly allocated to normal group (Group A) , model group (Group B), aminoguanidine (AG) group (Group C), NZP group (Group D) and NZP + AG group (Group E). Rat models of endotoxic shock were induced by intravenous injection of lipopolysaccharide (LPS) and intraperitoneal injection of galactosamine (D - Gal). Plasma NO level was detected by Griess method, the expression of inducible NOS (iNOS) and endothelial NOS (eNOS) was measured by immunohistochemical method and their activities were assayed by isotope labeling. Results] NZP decreased the plasma NO level and the activity and expression of iNOS in lung tissue, increased the expression of eNOS and reduced the lung injury. Conclusion] NZP can decrease the lung injury of rats with endotoxic shock and its mechanism may be related to the regulation of NOS expression in lung tissue.
