蛋白质-多糖复合纳米纤维膜用于皮肤缺损修复实验研究

摘    要:目的通过动物实验的方法评价由胶原蛋白-壳聚糖(两者比例为8:2)、明胶-透明质酸(两者比例为9:1)复合纳米纤维膜对于SD大鼠背部全层皮肤缺损创面修复的作用。方法用胶原蛋白-壳聚糖、明胶-透明质酸复合纳米纤维膜覆盖雌性SD大鼠皮肤缺损创面,两组各用大鼠15只;对照组仅覆盖油纱和纱布敷料,用大鼠15只。实验组和对照组大鼠背部皮肤缺损的面积均为2×1.5 cm2。分别于术后第3、7、14天观察SD大鼠大体情况、创面情况,并在第3、7、14天分别取创面组织行H-E染色对比观察。结果与对照组相比,实验组创面修复情况较对照组好,其中尤以胶原蛋白-壳聚糖复合生物材料的促创伤修复作用更明显。结论胶原蛋白-壳聚糖(8:2)复合纳米纤维膜较普通纱布敷料和明胶-透明质酸(9:1)复合纳米纤维膜能更好的促进创伤修复、愈合,有着较好的科研和临床应用前景。

Abstract:【Objective】 Animal experimental method was used to evaluate the role of Collogen-Chitosan(8:2) and Gelatin-hyaluronic acid(9:1) compound Nanofiber membrane to the SD rats full-thickness skin loss.【Methods】 Collogen-Chitosan(8:2)and Gelatin-hyaluronic acid(9:1) compound Nanofiber membrane were covered the back defect of the experimental group′s SD rats,while the control group was only covered oily waste and gauze;Each group using 15 rats;The area of skin loss are 2×1.5c㎡.The general survey of wound surface and H-E staining were done on 3th,7th,14th day respectively.【Results】 Compared with the control group,the tissue repair of the experimental groups is better,and especially,Collogen-Chitosan(8:2) compound Nanofiber membrane group is more effective.【Conclusion】 Collogen-Chitosan(8:2) compound Nano-biomaterial in respects of accelerated wounded skin repair is best,and has broad prospect in scientific research and clinical application.
