日本血吸虫成虫脲溶42 kD蛋白的分离及其初步诊断应用

目的 :分离日本血吸虫成虫脲溶抗原中有免疫学活性的抗原分子 ,探讨其诊断血吸虫病的效果。方法 :利用SDS PAGE及免疫印迹法分析日本血吸虫成虫脲溶抗原 ,采用电泳层析法分离有免疫活性的 4 2kD蛋白 ,ELISA分析 4 2kD蛋白诊断血吸虫病的灵敏度和特异性。结果 :电泳层析获得了具有免疫学活性的 4 2kD蛋白 ;分离的 4 2kD蛋白诊断血吸虫病的灵敏度和特异性分别为 88.0 9%和 97.0 6 %。结论 :日本血吸虫成虫脲溶 4 2kD蛋白是一种日本血吸虫的血清学抗原 ,可用于血吸虫病诊断

Fractionation of 42 kD Protein from Urea-soluble Schistosoma Japonicum Adult Worm Antigens and Its Primary Use for Schistosomiasis japonica Diagnosis

Objective To isolate proteins with good immunogencity from urea-soluble Schistosoma japonicum(Sj)adult worm antigens and evaluate its potency for schistosomiasis jaonica diagnosis. Methods Urea-soluble adult worm antigens from Sj were analysed by SDS-PAGE and immunoblotting and a 42 kD protein with good immunogencity was fractionated by electrophoretic chromatography. The fractionated 42 kD protein was used to detect IgG of schistosomiasis patients by ELISA. Results A 42 kD protein which reacted to rabbit sera infected with Sj was obtained by electrophoretic chromatography. The sensitivity and specificity of 42 kD protein for diagnosis of schistosomiasis japonica by ELISA were 88.09% and 97.06% respectively.Conclusion The 42 kD protein was obtained by electrophoretic chromatography and the protein was useful for diagnosis of schistosomiasis japonicum.
