生脉成骨方对股骨头坏死I型胶原表达的调节作用研究

目的　研究生脉成骨方对股骨头坏死Ⅰ型胶原mRNA表达的影响以揭示其作用机制。方法　以内毒素加甲基强的松注射制备兔骨坏死模型 ,分为模型组及生脉成骨方组 ,分批于用药后 4,8,12周处死 ,于原位杂交法测定骨Ⅰ型胶原表达 ,进行病理学检查 ,并与正常对照组比较。结果　模型组各时段成骨细胞及骨髓基质细胞内的Ⅰ型胶原mRNA的阳性表达减少 ;中药组 4周时mRNA的表达信号强度和量稍弱于正常组而稍高于模型组 ,8周和 12周时其信号强度和量以及阳性表达的骨髓基质细胞分布进一步改善 ,接近于正常组。结论　生脉成骨方在股骨头坏死的治疗中能促进成骨细胞功能活跃 ,增加骨髓基质细胞向前成骨细胞的分化 ,促进Ⅰ型胶原的合成

Regulatory effect of Shengmai Chenggu Recipe on collagen type I in necrotic femoral head

Objective It is to explore the effect of Shengmai Chenggu Recipe on collagen type I and its therapeutic mechanism in treatment of femur head necrosis (FHN). Methods Rabbit model of FHN was induced by injection of endotoxin and methylprednisolone. The model rabbits were divided into model group and herb group. They were sacrificed in batches in the 4th, 8th and 12th weeks after treatment, the femur head was taken for detecting expression of collagen type I mRNA with in situ hybridization and pathologic examination. All data were compared with that of control group. Results In situ hybridization indicated expression of type I collagen mRNA in osteoblasts and osteopregenitor cells decreased in model group. In herb group, at 4th week, the intensity and quantity were a little less than those of normal and slightly higher than those of model group, at 8th and 12th weeks they restored to normal and the positive bone marrow matrix cells also improved close to normal. Conclusion Shengmai Chenggu Recipe can promote osteoblasts proliferation and differentiation, thus to increase the synthesis of type I collagen.
