在体外培养中吡喹酮对日本血吸虫肝门型童虫活动及存活影响的观察

用体外培养技术观察了吡喹酮对日本血吸虫肝门型童虫活动及存活情况的影响。童虫接触较低浓度(1μg/ml以下)吡喹酮后表现为短时间兴奋,继而出现轻度挛缩,虫体活动减弱,换正常培养基后活动很快恢复并能正常存活;而接触较高浓度(1μg/ml以上)吡喹酮后,童虫立即出现强直性挛缩,活动几乎停止,短时间(6h内)换正常培养基童虫活力尚可恢复并能正常存活,延长与药物接触时间童虫活力不能恢复,继续培养时也不能存活。

An Observation of Praziquantel Effecting on the Activity and Survival of Hepatic-portal-phase Schistosomuiae of Schistosoma japonicum in vitro

The effect of praziquantel on the activity and survival of the hepatic- portal-phase schistosomulae of Schistosoma japonicum was investigated in vitro.Praziquantel at a concentration of 0.1μg/ml caused evident changes in the worm's activity.After the activity was increased for a short time,the schistosomulae in contact with the drug showed a slight contraction and the mobility of the worm remained at a low level.The activity of the juvenile could completely recover when they were transfered into drug-free media within 48h.The parasites in contact with 1.0μg/ml parziquantel displayed medium contraction after the occurrence of strong movement for a few seconds.The activity of the worm could recover when they transferred into drug-free media within 12h,The schistosomulae showed strong contrac- tion immediately when they touched the drug at concentration of 10 and 100μg/ml.The movement of the worms almost stopped after keeping touch with the drug at those concentrations.The activity of the parasites could recover when they were transferred into drug-free media within 6h.The report here is the preliminary observational results of prazquantel effecting on the activity and survival of S.japonicum schistosomulae in vitro.And also reported is the normal mobile type of the hepatic-portal-phase sehistosomulae in vitro.
