染铅早期大鼠海马各区NOS和nNOS阳性神经元的变化

摘    要:为了解海马各区NOS和nNOS阳性神经元在铅影响学习记忆神经毒机理中的作用 ,采用NADPH 黄递酶组织化学和免疫组化ABC法 ,观察了大鼠腹腔注射 130mg kg醋酸铅溶液染毒 3、6、12h和 2 4h后海马CA1 和CA3 区NOS和nNOS的阳性神经元数目的变化。结果显示 ,与对照组相比 ,大鼠染铅 6h时 ,海马CA1和CA3 区nNOS阳性神经元数目明显增加 (P <0 0 5 ) ;染铅 12h时 ,海马CA1 和CA3 区NOS和nNOS的阳性神经元数目均明显减少 (P <0 0 1) ;而染铅 2 4h时 ,只有海马CA1 区NOS和nNOS阳性神经元数目仍明显减少(P <0 0 5 ) ;而CA3 区未见明显变化。本研究提示海马不同亚区NOS和nNOS阳性神经元的时程变化可能与铅的神经毒作用有关

Abstract:In order to investigate the role of NOS and nNOS in the neurotoxic mechanism of lead(Pb)on learning and memory. NOS and nNOS positive neurons in CA 1 and CA 3 areas of hippocampal in the rats were observed by using NADPH-diaphorased(NADPH-d) histochemistry and immunohistochemistry after lead-intrapertoneal injection at the dosage of 130mg/kg body weight for 3, 6,12 and 24 hours respectively. Compared with the control, time dependent changes of NOS and nNOS positive neurons in the hippocampal CA 1 and CA 3 areas on lead exposed rats were as follow: CA 1 and CA 3 areas nNOS positive neurons were increased(P<0.05) after lead-exposed for 6h; but CA 1 and CA 3 areas NOS and nNOS positive neurons were decreased(P<0.01) after lead-exposed for 12h; after lead-exposed for 24h, only in CA 1 area, NOS and nNOS positive neurons were decreased(P<0.05). The results indicated that the time dependent changes of NOS and nNOS positive neurons in CA 1 and CA 3 areas of hippocampus may participate in the neurological toxicity of lead.
