摘    要:目的观察脱氢表雄酮(DHEA)诱导多囊卵巢综合征(PCOS)大鼠模型21 d、49 d后血清性激素水平和卵巢形态学改变,比较不同时长DHEA诱导PCOS大鼠模型的差异,为选择合适的诱导时长提供实验依据。方法将28只SPF级SD雌性大鼠分为正常组(n=10)和造模组(n=18),正常组大鼠每日皮下注射0.2 mL的注射用大豆油剂48天,造模组大鼠每日皮下注射脱氢表雄酮20天。于第11天采集并观察各组大鼠阴道脱落细胞;第21天,尾静脉采血检测各组血清睾酮(T)、卵泡刺激素(FSH)、黄体生成素(LH)水平,同时各组随机抽取2只大鼠进行卵巢组织HE染色观察,并将造模组再分为模型组Ⅰ(n=8)和模型组Ⅱ(n=8)。模型组Ⅰ每日皮下注射0.2 mL的注射用大豆油剂,模型组Ⅱ每日皮下注射脱氢表雄酮,均连续注射28 d。第49天,采集各组血清及卵巢标本,分别检测各组血清T、FSH及LH水平,HE染色法观察各组卵巢组织形态学变化。结果造模组大鼠动情周期失去规律;第21天,造模组血清T、LH水平高于正常组(P<0.01),FSH水平与正常组比较差异无统计学意义;第49天,模型组Ⅰ及模型组Ⅱ的T、LH水平高于正常组(P<0.01);模型组Ⅱ的T水平高于模型组Ⅰ(P<0.01);模型组Ⅰ及模型组Ⅱ的FSH水平和黄体个数均低于正常组(P<0.05,P<0.01),同时模型组Ⅱ黄体个数少于模型组Ⅰ(P<0.01);模型组Ⅰ卵巢指数高于正常组,模型组Ⅱ卵巢指数低于正常组,差异无统计学意义,模型组Ⅱ卵巢指数低于模型组Ⅰ(P<0.05)。结论采用DHEA诱导20 d的PCOS大鼠模型,经历4周后,不存在自愈,与持续造模相比更为简便易行,同时其卵巢形态结构以及激素水平变化也更符合临床表现。

Abstract:Objective:To observe the changes of serum sex hormone level and ovarian morphology of polycystic ovary syndrome(PCOS)rats induced by dehydroepiandrosterone(DHEA)after 21 days and 49 days,and to compare the differences of PCOS rats induced by different length of DHEA at the same time,so as to provide experimental basis for selecting the appropriate induction time.Methods:Twenty-eight SPF-class SD female rats were divided into normal group(n=10)and model group(n=18).Rats in normal group were injected subcutaneously with 0.2 mL of injectable soybean oil for 48 days.Model group rats were injected subcutaneously with DHEA for 20 days.On the 11th day,the vaginal exfoliated cells of each group were collected and observed.On the 21st day,blood samples were taken from the tail vein to detect the levels of serum testosterone(T),follicle stimulating hormone(FSH)and luteinizing hormone(LH)in each group.Two rats were taken for HE staining of ovarian tissue,and the model group was subdivided into model groupⅠ(n=8)and model groupⅡ(n=8).Model groupⅠwere injected subcutaneously with 0.2 mL of injectable soybean oil for 28 days,and model groupⅡwas injected subcutaneously with DHEA for 28 days.On the 49th day,serum and ovarian specimens were collected and serum T,FSH and LH levels were measured.HE staining was used to observe the morphological changes of ovarian tissue.Results:The estrous cycle of the rats in the model group was lost.On the 21st day,the serum T and LH levels of the model group were higher than those of the normal group(P<0.01).There was no significant difference in the FSH level between the model group and the normal group.On the 49th day,The T and LH levels in the model groupⅠand model groupⅡwere higher than those in the normal group(P<0.01).The T level in model groupⅡwas higher than that in model groupⅠ(P<0.01).The FSH level and corpus luteum in model groupⅠand model groupⅡwere higher than.The number of the corpus luteum was lower than that of the normal group(P<0.05,P<0.01),while the number of corpus luteum in the model group was lower than that of the model groupⅠ(P<0.01).The ovarian index of the model group I was higher than that of the normal group.There was no statistically significant difference between the two groups(P<0.05).Conclusion:After 4 weeks,there was no self-healing in the PCOS rat model induced by DHEA for 20 days.Compared with the continuous model,it was easier and easier to establish,and the changes of ovarian morphology and hormone level were more consistent with the clinical manifestations.
