苁蓉舒痉颗粒中间体总多糖的质量控制研究

摘    要:目的建立分光光度比色法测定苁蓉舒痉颗粒中间体的总多糖含量。方法采用蒽酮-硫酸比色法,以无水葡萄糖为对照,在582 nm波长处测定总多糖含量,进行精密度、稳定性、重复性及加样回收率等方法学考察。结果无水葡萄糖质量浓度在3.41~27.26μg/mL范围内吸光度值呈良好的线性关系,回归方程为A=3.73×10^2C+4.47×10^2(r=0.998 8),平均回收率为98.32%(RSD=1.65%),3批苁蓉舒痉颗粒中间体的总多糖含量分别为20.48、22.04、24.81 mg/g。结论方法准确,重复性良好,可用于苁蓉舒痉颗粒中间体总多糖质量控制。

Abstract:Objective:To establish a visible spectrophotometric colorimetric method for the determination of total polysaccharides content in Congrong Shujing granules intermediate.Methods:The anthrone-sulfuric acid colorimetry was used,with anhydrous glucose as the control,to determine the total polysaccharides content at a wavelength of 582 nm.The precision,stability,repeatability and recovery rate of the sample were evaluated for this method.Results:A good linear relationship was observed when the concentration of anhydrous glucose was in the range from 3.41 to 27.26 μg/mL.The regression equation was A=3.73×10^2 C+4.47×10^2(r=0.998 8),and the average recovery rate was 98.32%(RSD=1.65%).The total polysaccharides contents of the three batches of Congrong Shujing granules intermediate were 20.48,22.04,and 24.81 mg/g respectively.Conclusion:The method can be used as a quality control of total polysaccharides in Congrong Shujing granules intermediate due to its good accuracy and repeatability.
