阿司匹林对STZ糖尿病大鼠外周血白细胞iNOS mRNA和胰岛细胞iNOS表达的影响

摘    要:目的:探讨早期应用阿司匹林对糖尿病大鼠外周血白细胞(PBL)iNOS mRNA、胰岛细胞iNOS表达的影响。方法:腹腔注射链脲佐菌素(streptozotocin,STZ)制备实验性STZ糖尿病大鼠模型,治疗组给予肠溶阿司匹林灌胃。检测血糖、血浆胰岛素含量、胰岛β细胞胰岛素的表达情况,及胰岛中iNOS、外周血白细胞iNOS mRNA阳性细胞率的变化。结果:治疗1月后.胰岛iNOS阳性细胞及外周血白细胞iNOS mRNA阳性细胞表达率明显降低(P〈0.05).结论:早期应用阿司匹林能明显降低外周血白细胞iNOS mRNA和胰岛中iNOS阳性细胞表达率,降低血糖水平,改善胰岛β细胞的分泌功能。

Abstract:Objective: To investigate the effects of aspirin on the expressions of leukocyte iNOSmRNA and islet cell iNOS in diabetic mellitus rats. Methods: Wistar diabetic rats were induced by streptozotocin(STZ) and divided into control and aspirin-treated groups at random. The levels of blood sugar and insulin were tested, and the alterations of expression of leukocyte iNOS mRNA and the expression of iNOS in islet cells were detected by hybridization in situ and immunohistochemistry. Results: The blood insulin increased and the blood sugar decreased obviously in aspirin-treated group. The percent of iNOSmRNA-positive leukocytes and iNOS-positive islet cells decreased obviously. Conclusion: Early use of aspirin can reduce the quantity of iNOSmRNA-positive leukocyte and iNOS-positive islet cells obviously.
