抗TNF-α单克隆抗体对VMC小鼠影响的实验研究

摘    要:目的:了解肿瘤坏死因子α(TNFα)在病毒性心肌炎小鼠发病中的作用及抗TNFα单克隆抗体对心肌损伤的保护效应。方法:BalB/c小鼠随机分为正常对照组、病毒感染组和病毒感染加抗体治疗组,测定感染后7d和14d心肌组织TNFα的表达、病理变化、超微结构改变和小鼠生存率。结果:①治疗组TNFα表达较感染组低(P<0.05);②治疗组心肌病理变化和超微结构改变减轻,生存率提高;③感染组TNFα表达与心肌病变积分呈正相关(P<0.05)。结论:TNFα在病毒性心肌炎发病中发挥重要作用,抗TNFα单克隆抗体对受损心肌有保护作用。

Abstract:Objective: To study the role of TNF-α in the development of viral myocarditis and the protective effect of anti-TNF-alpha monoclonal antibody on myocardial injury in mice. Methods: Male BalB/c mice were randomly divided into the control group, infection group, infection with antibody treatment group. The expression of TNF-α, the pathologic changes, the ultrastructure of heart and the survival rate were studied at the 7th and 14th day after infection. Results: ① Compared to that in infection group, the expression of TNF-α reduced in treatment group(P<0.05);② The changes of ultrastructure and pathology reduced, the survival rate increased in treatment group;③ There was a positive correlation between the expression of TNF-α and the changes of myocardial histopathologic scores in infection group (P<0.05). Conclusion: TNF-α might play an important role in the pathogenesis of viral myocarditis, and anti-TNF-α monoclonal antibody could reduce myocardial injuries and protect myocardial tissues.
