标本前处理方法对乳汁HBV DNA检测的影响

目的探索一种简单易行的乳汁标本前处理方法 ,以便准确测定乳汁中的HBVDNA浓度。方法 4℃冰箱过夜后取中间层乳汁进行检测,以离心后的上清液、上清液浓缩、沉淀3种方法平行检测20份大三阳产妇的乳汁HBVDNA含量。结果沉淀方法检测阳性率为98%,明显高于其他2种方法 (均P0.01)。阳性标本HBVDNA的对数浓度为4.8±0.5,与其他2种方法比较,差异有统计学意义(均P0.01)。结论 4℃冰箱过夜后取中间层乳汁离心沉淀检测HBVDNA是比较准确可靠的,方法简单易行,值得临床推广应用。

Effect of different sample pretreatment methods on HBV-DNA quantitation in breast milk

Objective To explore an easy and reliable sample pretreatment method of breast milk for effective HBV-DNA quantitation.Methods 20 samples were collected from parturients with serum HBsAg HBeAg HBcAb positive.The samples were centrifugated after refrigeration overnight at 4 ℃.The supernatant,concentrated supernatant,precipitation of each sample were used as samples to detect HBV DNA by fluorescent quantitative PCR.Results HBV DNA positive ratio and log concentration of precipitation were 98% and 4.8±0.5 respectively,which were significantly higher than those of supernatant,concentrated supernatant（P〈0.01）.Conclusion It is suggested to take precipitation of centrifugated breast milk after refrigerating overnight at 4 ℃ for HBV-DNA quantitation.
