蜂胶总黄酮对大鼠心肌缺血再灌注损伤Fas、Bax和Bcl-2基因蛋白表达的影响

摘    要:目的研究蜂胶总黄酮对大鼠心肌缺血再灌注损伤相关基因蛋白表达的影响。方法建立大鼠心肌缺血再灌注损伤模型,研究Fas、Bax、Bcl2、p53基因蛋白表达。结果缺血30min,再灌注48h后,模型组和蜂胶总黄酮组的大鼠心肌Fas基因蛋白表达均明显高于假手术组,差异有显著性;且蜂胶总黄酮组的Fas表达均低于模型组,证明蜂胶总黄酮对Fas表达具有明显的抑制作用。Bax基因的蛋白表达:模型组与蜂胶总黄酮组均高于假手术组,但蜂胶总黄酮组与模型组之间并无差异。Bcl2基因蛋白的表达:模型组与蜂胶总黄酮组均高于假手术组,其中蜂胶总黄酮高剂量组与假手术组比较差异有显著性,而其它各组间比较差异无显著性。p53基因蛋白表达:各组切片经免疫组化反应后,均未见明显的p53基因蛋白表达迹象。结论蜂胶总黄酮通过抑制Fas蛋白表达,上调Bcl2蛋白表达而减少了心肌细胞的凋亡,保护了心肌组织的结构与功能。

Abstract:Aim To study effects of the total flavones of propolis(TFP)on Fas、Bax and Bcl-2 genes protein expression in myocardial ischemic-reperfused injury rats. Methods the model of myocardial ischemic-reperfused injury in rats was established. Fas、Bax and Bcl-2 genes protein expression were detected in myocardial ischemic-reperfused injury rats. Results Following ischemia 30min and reperfusion 48 h,Fas gene protein expression in rats myocardium in ischemia reperfusion group and TFP group significantly increased compared with that in sham-operation group,and Fas gene protein expression in every medicine pretreatment group was significantly lower than that in ischemia reperfusion group. Bax gene protein expression in ischemia reperfusion group and TFP group were both significantly higher than that in sham-operation group, but there was not significant difference between the TFP group and ischemia reperfusion group. Bcl-2 gene protein expression in ischemia reperfusion group and TFP group were both significantly higher than that in sham-operation group, and the expression in TFP(200 mg·kg -1 ) group significantly increases compared with sham-operation group. There was no sign of p53 gene protein expression in all sections by S-P immunohistochemistry staining in our experiment.Conclusion TFP can inhibit Fas gene protein expression, at the same time induce Bcl-2 gene protein expression to inhibit myocardial cell apoptosis.
