人、小鼠和大鼠的ADRB3比较及其生物学和药理学意义

摘    要:目的:比较人、小鼠和大鼠的ADRB3并讨论结果的生物学和药理学意义。方法:由NCBI的GemBank数据库,搜索出人、小鼠和大鼠的ADRB3的基因序列,经过对准和比较。计算出其氨某酸序列的相似性和差异。结果:人、小鼠和大鼠的ADRB3的氨基酸序列长度分别为:408、404、400aa;人和小鼠、人和大鼠、小鼠和大鼠的ADRB3的氨基酸序列的排列和组成相似性分别为:79.3%、78.2%、92.6%;差异分别为:20.7%、21.8%、7.4%。最大差异在第四胞内段羧基尾部。结论:1.人与小鼠的序列长度更接近,排列和组成的差异更小。2.从序列的长度和相似性看。人与小鼠更接近,故推断进化顺序应为。3.序列最大差异在与胞内信号转导有关的第四胞内段。

Abstract:Objective: To compare the beta-3 adrenoceptor of human with the beta-3 adrenoceptor of mice and rats, and discuss the biological and pharmacological implications of the comparative results. Methods: The genomic sequences of beta-3 adrenoceptors of human, mice and rat were searched out from the GenBank of NCBI, and the similarities and differences of their amino acid sequences were computed out by aligning and comparing. Results: The lengths of amino acid sequences of the beta-3 adrenoceptors in human, mice and rats are 408,404 and 400 aa; The similarities and differences in the permutation(or order) and component among amino acid sequences of the beta-3 adrenoceptors of human, mice and rat were 79.3%, 78.2%, 92.6% and 20.7%, 21.8%, 7.4%, respectively. The most differences lie in the carboxyl tail in the 4th intracellular segment. Conclusions: 1. The sequence length between amino acid of the beta-3 adrenoceptors of human and mice was more close than that between human and rat, and the differences in the permutation(or order) and component among amino acid sequences of the beta-3 adrenoceptors of human and mice was less than of human and rat. 2. As far as the sequence length and the similarity go, the human is more close to the mice. 3. The Most difference of sequences was in the 4th intracellular segment associated with intracellular signal transduction.
