Sirt1在胃癌及癌旁组织中的表达差异研究

目的应用寡聚核苷酸基因表达谱芯片研究胃癌组织与癌旁组织的基因表达谱差异。方法应用包含44，000条人类全长基因的寡聚核苷酸芯片检测胃癌组织与癌旁组织，并比较存在两者之间存在明显上、下调的表达差异基因，随后用RT—PCR方法进行验证分析。结果基因芯片筛选出存在差异表达的基因，Siftl基因的表达水平在胃癌组织均存在明显下调，下降至对应癌旁组织的（0．318±0．032）～（0．169±0．013）倍不等（t=-3．374，P〈0．05）。经RT—PCR对胃癌组织与癌旁组织中Sift1基因的表达水平进行比较，发现胃癌组织中的实时荧光定量PCR的比值为0．258（X2=5．20，P〈0．05），与基因芯片的结果一致。结论Sirt1在胃癌组织与癌旁组织中的表达存在明显差异，Sirt1基因的表达可能与胃癌的发生发展密切相关。

Expression of Sirt1 in tissues of patients with gastric carcinoma

Objective To explore the changes in gene expression in tissues of patients with gastric carcinoma by cDNA microarray. Methods A 44,000 gene cDNA microarray （Affymetrix） was used to examine gene expression in the tissues of patients with gastric carcinoma. The Sirtl genes were identified and subjected to realtime PCR analysis. Results The expression of Sirtl in tissues of gastric carcinoma （0. 318 ±0. 032） ~ （0. 169 ±0. 013） time] was extremely lower than in tissues of paragastric carcinoma （ 1 time）. The results of Real-time PCR were consistent with those of genechip analysis. Conclusions The expression of Sirtl was obviously different between tissues of gastric carcinoma and para - gastric carcinoma. Sirtl may be associated with carcinogenesis of HCC.
