加味四君子汤诱导小鼠肿瘤细胞凋亡的实验研究

目的:从诱导肿瘤细胞凋亡的角度探讨中药复方加味四君子汤抗肿瘤作用的机理。方法:1.运用透射电镜观察肿瘤组织中凋亡细胞的超微结构。2.运用原位末端标记法观察凋亡细胞形态并计数,计算凋亡指数。3.运用流式细胞仪分析加味四君子汤对肿瘤细胞增殖周期的影响。结果:1.透射电镜观察到凋亡细胞超微结构发生了一系列典型变化。2.原位末端标记法镜下可见不同时期的凋亡细胞,加味四君子汤组凋亡指数为12.29%,与生理盐水组比较差异非常显著(P<0.01)。3.流式细胞仪分析发现加味四君子汤组G0-G1期细胞增加,S期和G2-M期细胞减少,中药组处于G0-G1期的细胞占57.88%,与生理盐水组比较差异非常显著(P<0.01)。结论:加味四君子汤抗肿瘤作用机理之一可能是将细胞周期阻滞在G0-G1期,继而导致受阻滞的细胞发生凋亡。

Iducing Tumor Cells Apoptosis of Jiawei Sijunzi Decoction

Objective:To study the mechanism of the anticancer activity of Jiawei Sijunzi Decoction(JSD) from its inducing tumor cells apoptosis point of view.Methods:1.To observe the ultrastructure of apoptotic cells in tumor tissue by electronic microscope.2.To observe the feature of apoptotic cell and acculate the apoptotic index(AI) by in situ end-labeling(ISEL) of fragmented DNA.3.To analyse the JSD′s inhibitory effect on tumor cell′s cycle by flow cytometry.Results:1.A series of ultrastructural changes were found.2.By the method of ISEL,we found the apoptotic cells of every phase. The AI of JSD group was 12.29 %(P<0.01).3.It was found that JSD arrested 57.88 %cells from G0-G1 to S phase by flow cytometry.Conclusion:One of the anticancer activity of JSD might be due to its effect of inducing tumor cells apoptosis by arresting cell′s cycle from G0-G1 to S phase.
