摘    要:研究人类端粒酶催化亚单位(hTERT)基因的反义寡核苷酸对宫颈癌细胞的影响。方法:采用TRAP-ELISA检测Hela细胞在反义核酸处理前后端粒酶活性的变化。结果:hTERT基因反义核酸能有效抑制Hela细胞端粒酶活性。结论:hTERT基因反义核酸能显著抑制肿瘤细胞的端粒酶活性,为肿瘤治疗提供了一个有效的新途径。

Abstract:Objective: To study the effects of antisense oligonucleotide against human telomerase catalytic subunit mRNA(hTERT mRNA)on human telomerase activity in cervical cancer cells in vitro. Method :Telomerase activity of cultured cervical cancer cells(Hela cells) was measured using the TRAP-ELISA assay before and after antisense treatment. Result: The antisense treatment could effectively inhibite the telomerase activity of Hela cells. Conclusion: Antisense oligonucleotide direct against hTERT mRNA could effectively inhibite the telomerase activity of tumor cells, which show an effective and novel approach for treatment of cancers.
