鸦胆子油对肝癌细胞SMMC-7721增殖的抑制作用

摘    要:目的:探讨鸦胆子油抑制肝癌细胞生长的机制。方法:人肝癌细胞SMMC-7721培养于含10%胎牛血清、100 U·ml^-1青霉素、链霉素的RPMI1640培养液中。以不同浓度的鸦胆子油乳加入培养液,作用一定的时间,进行MTT比色实验,检测生长抑制率及分析细胞周期。结果:MTT比色法测定显示鸦胆子油抑制肝癌细胞的生长,随着浓度增加和作用时间的延长,抑制作用呈现逐渐增加的趋势。细胞周期分析提示:G0-G1期比例逐渐增加,而S期比例逐渐减少,1μg·ml^-1组中各时间点及2μg.ml-1组中各时间点均有显著差异(P〈0.01)。G2-M期细胞比例减少,但差异无统计学意义(P〉0.05)。结论:鸦胆子油抑制体外培养的人肝癌细胞的增殖,机制可能是将肝癌细胞阻滞于G0-G1期。

Abstract:Objective:To investigate the inhibition effect of Brucea javanica oil emulsion on the proliferation of human liver cancer cells.Methods: The liver cancer cells SMMC-7721 were cultivated with Brucea javanica oil emulsion.The effects of Brucea javanica oil emulsion on the proliferation of SMMC-7721 cells were observed using MTT method and flow cytometry.Results: Brucea javanica oil emulsion could inhibit the growth of SMMC-7721 cells in dose-dependent and time-dependent manner.The cell cycle analysis showed that the percentage of G0/G1 phase cells increased and S phase cells decreased.There was significant deviation(P〈0.01) in interclass of the concentration of Brucea javanica oil emulsion 1μg·ml^-1 and 2μg·ml^-1(P〈0.01).The percentage of G2/ M phase cells decreased but there wasn' t significant deviation(P〉0.05).Conclusion: Brucea javanica oil emulsion could inhibit the proliferation of SMMC-7721 cells in vitro and the mechanism may relate to its preventing the cell cycle in G0/G1 phase.
