Abstract:Objective To explore the effect of bioactive components in mulberry leaves,including mulberry leaves extracts(MLE),mulberry leaves polysaccharides(MLP) and mulberry leaves flavonoids(MLF),on glucose metabolism in insulin-resistant HepG2 cells.Methods The insulin resistant models of HepG2 cells were induced by high concentration of insulin for 24 h.The glucose consumption of cells,the liver glycogen,hexokinase(HK) and pyruvate kinase(PK) were measured after incubation with bioactive components in mulberry leaves.Results MLE and MLF advanced glucose uptake in insulin-resistant HepG2 cells.The liver glycogen synthesis,HK secretion and PK secretion increased significantly after incubation with MLE.Conclusion MLE and MLF can improve the glucose consumptions in insulin-resistant model of HepG2 cells.MLE can enhance HK and PK vitality,promote blood glucose to enter the liver cells and increase glycogen synthesis.MLE may regulate the glucose metabolism and improve the symptoms in diabetes mellitus.
