Abstract:Objective To establish a HPLC method to determine uridine and adenosine in Ophiocordyceps sinensis from the main producing areas of Tibet. Methods Uridine and adenosine were separated on Agilent C18 column (250 mm × 4.6 mm,5 μm) and detected at 260 nm. The mobile phase was consisted of methanol and water using a gradient elution. The column temperature was 33 ℃ and the flow rate was 1.0 mL/min. Results The linearity of uridine and adenosine were 32.95 - 439.3 μg/mL ( r = 0. 999 9 ) and 3. 486 -46.48 μg/mL(r =0. 999 9) ,and the average recovery ratios were 98.5% and 95.6%. The RSD were 1.8% and 2.4% ,respectively. The contents of uridine and adenosine ranged from 1. 209 to 2. 192 mg/g and 0.083 8 to 0. 884 mg/g respectively. Conclusion The method is simple and accurate, and it can be used for the determination of uridine and adenosine quickly. It provided a reference to control the quality of Ophiocordyceps sinensis from Tibet.
