摘 要: | 目的:观察补肾填精方对γ射线所致辐射损伤小鼠模型的作用,并通过观察其对DNA损伤修复ATM/ATR通路的影响,探讨该方抗辐射的作用机制。方法:将雌雄各半的balb/c小鼠,分成正常组、病理组和补肾组。采用γ射线损伤小鼠模型,在造模后取脾脏运用real-time RT-PCR方法检测该通路关键基因ATM和ATR基因的表达,并应用免疫组化方法检测ATM和ATR蛋白表达,结合图像分析技术进行分析。结果:补肾组ATR基因表达与病理组相比,有显著差异(P<0.05),而ATM基因表达在各组中的差异不明显;免疫组化结果与RT-PCR结果一致。结论:补肾填精方可促进ATR基因和蛋白的表达增强。

Abstract: | Objective:To discuss the mechanism of Bushen Tianjing Fang,we observed the role of Bushen Tianjing Fang to mice models damaged byγray,and its role in DNA damage repair ATM/ATR pathway.Methods:The male and female balb/c mice were divided into normal group,the pathological control group and the kidney group.Ray irradiated mice with γ model were used.By using real-time RT-PCR method,the gene expression of the key genes ATM and ATR were detected and the proteins were analyzed by image analysis Technology for analysis.Results:Compared with the pathology group,the expression of ATR in kidney group,there are significant differences(P<0.05),while the ATM gene expression in each group were not significantly different.The ATR protein expressions in the kidney group were significantly different compared with the pathology group.Conclusion:Bushen Tianjing Fang can enhance ATR gene and protein expression. |
