巢蛋白nestin在人甲状腺常见肿瘤组织的表达

目的探讨nestin在人甲状腺常见肿瘤及非肿瘤组织中的表达及意义。方法回顾性分析25例甲状腺乳头状癌、25例甲状腺腺瘤的病理资料,与同期收治的19例结节性甲状腺肿及12例甲状腺腺瘤瘤旁正常甲状腺组织比较。采用免疫组织化学（IHC）EnVision法检测其nestin蛋白的表达,应用激光捕获显微切割技术（LCM）RT-PCR分析加以验证。结果 IHC结果显示,nestin在甲状腺乳头状癌和甲状腺腺瘤组织中阳性表达率分别为56.0%（14/25）和32.0%（8/25）,两组间的表达率差异无统计学意义。若将肿瘤组（44.0%,22/50）分别与结节性甲状腺肿（0/19）和瘤旁正常甲状腺组织（0/12）比较,差异均具有统计学意义（χ2=14.267,P=0.000）。LCM后RT-PCR分析结果与IHC结果一致。结论 nestin在甲状腺常见肿瘤组织高表达而在非肿瘤性甲状腺组织不表达,提示nestin在甲状腺常见肿瘤的发生中可能起重要作用。

Nestin expression frequently occurs in human thyroid tumors

Objective To explore nestin expression in human thyroid neoplasms and non-tumorous thyroid tissues.Methods Eighty-one thyroid tissue blocks from 25 patients with papillary carcinoma（PTC）,25 patients with adenoma,19 patients with nodular goiter and 12 normal thyroid tissues from surrounding tissue of surgically removed adenoma were used to detect nestin protein expression by immunohistochemistry（IHC） technique and nestin RNA expression by laser capture microdissection（LCM） technique and RT-PCR.Results The positive signal of IHC was mostly from follicular epithelial cells in papillary carcinoma and adenoma tissues.The detected positive rates of nestin in papillary carcinoma and adenoma tissues were 56.0%（14/25） and 32.0%（8/25）,respectively.In both multinodular goiters and normal tissues,the positive rates were 0.Statistical analysis indicated that nestin expression was significantly different between the non-tumorous thyroid tissues and neoplasms（χ2=14.267,P=0.000）.Conclusions Immunohistochemical staining for nestin may be useful for the differential diagnosis between the common thyroid tumors and multinodular goiter and normal tissues but the mechanism of nestin expression frequently in thyroid tumors remains unclear.
