Graves 病患者外周血淋巴细胞亚群及其增殖反应的检测分析

采用单克隆抗体间接免疫荧光法检测40例不同病期的 Graves 病(GD)患者外周血淋巴细胞亚群。结果表明:GD 患者发病期,外周血 CD_3~+、CD_8~+细胞百分率均明显低于正常人(P<0.05,<0.01),CD_4~+/CD_8~+比值和 B 细胞百分率明显增高(P<0.01);具有 GD 家族史的缓解病人,异常持续存在,而无 GD 家族史的缓解病人 T 淋巴细胞亚群恢复正常。此外,GD 患者外周血单个核细胞对PHA 增殖应答能力低下。说明 GD 患者外周血淋巴细胞数目和功能均存在异常。提示患者的细胞免疫和体液免疫改变与发病机理有关。

Studies on Lymphocyte Subsets and Blastogenic Response in Patients with Graves Diseases

The T-lymphocyte subsets and B lymphocytes in the peripheral blood of 40patients with Graves diseases and healthy volunteers were determined with monoclonal anti-bodies (McAbs) by indirect fluorescence assay (IFA).The lymphocyte proliferative re-sponse to phytohemagglutinin (PHA) was investigated by ~3H-TdR incorporation assay.Theresults showed that the percentage of CD_3~+ cells and CD_8~+ cells of patienets with Gravesdiseases in active stage significantly decreased,and the ratio of CD_4~+/CD_8~+ and the percentageof B lymphocytes markedly increased as compared with the control group.During remissionperiod,the abnormalities in T-lymphocyte subsets still persisted in the patients with Gravesdisease family history and returned to normal in the patients without Graves disease familyhistory.Blastogenic response of peripheral blood mononuclear cell's (PBMC) to PHA wassignificantly diminished in patients with Graves diseases as compared with that in the healthyvolunteers (P<0.01).The above findings suggest that the pathogenic mechanism of theGraves diseases is related to abnormality in cellular and humoral immunity in patients withGraves diseases.
