AGL提取物总黄酮的部分药效学研究

摘    要:目的:探讨碱-酸法提取的AGL总黄酮的毒性及其对SD大鼠血压及兔动脉粥样硬化动物模型的影响。方法:(1)选用SD大鼠,静脉推注AGL总黄酮(1mg/kg,2mg/kg),观察血压波动变化。(2)选用新西兰兔,高脂饲料喂养形成动脉粥样硬化动物模型,灌胃给予AGI。总黄酮(5mg/kg),观察血清总胆固醇(TC)、低密度脂蛋白-胆固醇(LDL-C)、高密度脂蛋白~胆固醇(HDL-C)、甘油三酯(TG)、C反应蛋白(CRP)及主动脉的病理学变化。结果:(1)小鼠对AGI。总黄酮最大耐受剂量为6g/kg。(2)AGL总黄酮(1mg/kg,2mg/kg)对SD大鼠血压的影响差异无统计学意义(P〉0.05)。(3)AGL总黄酮(5mg/kg)对高脂饲料喂养形成的兔动脉粥样硬化动物模型的影响:AGL总黄酮组血清TC、LDL、HDL水平与模型组比较,差异无统计学意义(P〉0.05);AGL总黄酮组血清TG、CRP水平虽有升高,但与模型组比较差异无统计学意义(P〉0.05)。病理学检查结果表明,主动脉内膜可见泡沫细胞、粉红色无定性物质。结论:用碱-酸法提取的AGL总黄酮对大鼠血压和动脉粥样硬化动物模型无显著影响。

Abstract:Objective: To investigate the acute toxicity of AGL flavonoids(extracted by the basic-acid method) and its effect on blood pressure and arteriosclerosis animal models.Methods:(1) Changes in blood pressure were observed by intravenous injections of AGL flavonoids(1 mg/kg, 2 mg/kg) in rats.(2) Experimental arteriosclerosis animal model were established in New Zealand albino rabbits by hyperlipoidal food,then AGL flavonoids were(5 mg/kg) given orally;Blood samples were collected to assess serum TC,LDL, TG,HDL,CRP and dimension of aortic changes in the histopathology.Results:(1)AGL flavonoids' maximum tolerance dose was 6 g/kg.(2) AGL Flavonoids showed no significant changes on blood pressure(P>0.05).(3) Compared with the hyperlipoidal food group,AGL flavonoids had no significant change in serum TC,LDL,HDL,but serum TG,CRP is high(P>0.05).Foam cells and atherosclerotic plaques were observed in aortic intima in the AGL flavonoids group.Conclusions: It is concluded that AGL flavonoids extracted by the basic-acid method possess no marked effects on blood pressure and arteriosclerosis models in rabbit.
