摘    要:目的:提取并纯化正常人脑脊髓的髓磷脂碱性蛋白(MBP),鉴定其活性,并用其诱导实验性自身免疫性脑脊髓炎(EAE)大鼠模型。方法:以人脑脊髓为原料,通过去脂、酸溶解、CM—52柱层析等步骤提取人MBP,鉴定其纯度后在Wistar大鼠诱导EAE。结果:所提取的MBP具有纯度高、免疫话性好的特点,与抗MBP抗体能特异性结合,并可成功地诱导出EAE动物模型,病理学证实发病大鼠的脑脊髓具有典型的脱髓鞘改变和胶质结节形成。结论:用人MBP在不加百日咳杆菌的条件下,同样可以诱导出EAE,这种动物模型体内的病理学改变与人类的多发性硬化症(MS)更具有相似性,适用于对MS的研究。

Abstract:Objective: To induce a meliorative model of experimental allergic encephalomyelitis (EAE). Methods: Purified human myelin basic protein was obtained by means of defatting with methanol-chloroform, acid extraction and CM-52 chromatography. After being identified, it was used to produce the model of EAE. Results: Human MBP purified by our lab, with good purification and im-munological activation, can be combined with the human MBP antibody. It can induce EAE in wistar rat, which showed large de-myelinated areas in the brain and spinal cord like human with multiple sclerosis (MS). Conclusion: The animal model of EAE induced by human MBP without BPV bears a resemblance to the features of MS, which promises to be a better model than ever before for the study of multiple sclerosis.
